摘要
目的调查贵州省侗族健康随机献血人群人类血小板抗原(humanplateletantigen,HPA)1—5系统基因多态性分布特点。方法采用聚合酶链一序列特异性引物技术(PCR.SSP)对贵州省无血缘关系侗族随机献血人154例血样进行HPA.1—5系统基因分型。结果贵州省黔东南州侗族HPA.1a、2a、3a、4a、5a、基因频率分别是0.942、0.741、0.926、0.954、0.773,HPA-lb、2b、3b、4b、5b、基因频率分别是0.058、0.286、0.084、0.045、0.227;经/检验,结果符合Hardy-Weinberg遗传平衡定律。随机输血HPA不配合的发生率依次为IHPA-4:49.11%,HPA-2:32.%,HPA-5:28.94%,HPA-3:14.35%,HPA-1:10.33%。与其他研究比较:贵州省侗族人群HPA-l-5系统基因频率与不同人群相比较:HPA-1与中国汉族、海南黎族、新疆维吾尔族相比较差异有统计学意义(P〈0.05);HPA-2与贵州省黔东南州苗族、中国汉族、新疆柯尔克孜族、海南黎族、西藏藏族、浙江畲族、美国白人相比较均差异有统计学意义(P〈0.05);HPA-3、4与贵州省黔东南州苗族、中国汉族、新疆维吾尔族、新疆柯尔克孜族、海南黎族、西藏藏族、浙江畲族、广西瑶族、美国黑入、美国白人相比较差异有统计学意义(P〈0.05);HPA-5-7贵州省黔东南州苗族、中国汉族、新疆柯尔克孜族、海南黎族、渐江畲族、广西瑶族、美国白人相比较均差异有统计学意义(P〈0.05)。结论贵州省侗族献血人群HPA-1~5系统基因具有独特的分布频率和群体结构特点,且不配合率较高,应建立本地区的血小板献血者基因分型数据库。
Objective To investigate the allele polymorphism and characteristic of HPA-1 5 genotyping of Dong ethnic group in Guizhou province, and to evaluate the HPA mismatched ratio between donors and recipients in randomly chosen ca- ses of blood transfusion, in order to investigate the platelet transfusion refractoriness in Li population. Methods Samples from unrelated platelet donors in Guizhou Dong population were genotyped with sequence specific primers (PCR-SSP) Results Guizhou province Qiandongnan Dong HPA-la,2a,3A,4a,5a,gene frequencies are 0. 942,0. 741,0. 926,0. 954, O. 773, HPA- 1 b,2b,3b, 4b, 5b, gene frequencies are 0. 0580. 286,0. 084,0. 045,0. 227 ;X2 test results in Hardy-Weinberg genetic equilibrium. The incidence of random blood transfusion HPA mismatch was as follows : HPA-4 :,'9. 11% , HPA-2 : 32% ,HPA-5:28. 94%, HPA-3 : 14. 35% , HPA-I : 10. 33%. Compared with other studies : Guizhou province, the Dong the crowd HPA-1 5 gene frequencies compared with different populations:the HPA-1 with the Han Chinese, Hainan Li ,Xinjiang Uygur compares the difference was statistically significant (P 〈 0. 05) ;the HPA -2 and Qiandongnan Miao of Guizhou Prov- ince, Han China, Xinjiang, Kirgiz, Hainan Li ethnic Tibetans in Tibet, Xijiang Shes white American compared differences were statistically significant (P 〈 0. 05 ) ; HPA-3,4 and Guizhou Province the Qiandongnan Miao,Chinese Hart, Uygur and Kirgiz in Xinjiang, Hainan Li ethnic Tibetans in Tibet,Xijiang Shes Guangxi Yao, African Americans,white Americans compared the difference was statistically significant (P 〈 0. 05 ) ; HPA-5 Qiandongnan Miao of Guizhou Province, Han Chinese, the Kirgiz in Xinjian.g, Hainan Li Xijiang Shes the Yao, white Americans compared differences were statistically significant ( P 〈 0. 05 ) Conclusion Guizhou province Dong blood donors HPA-1 5 system gene has a unique distribution characteristics of fre-quency and population structure, and not with the higher rate, should be established in the region platelet donors genotyping da- tabase.
