高效液相色谱法测定丙戊酸血浓度

摘要 本文建立了采用BrMMC将丙戊酸转化为可紫外检测的酯,再经RP-HPLC测定的丙戌酸血浓度测定方法.在15-200μg/ml范围内,峰高比与浓度呈良好的线性关系,r=0.9991;平均回收率为101.2%.方法准确、灵敏、选择性强、重现性好,操作简便快速.应用于34例患者的治疗药物监测,取得良好的临床效果,本法适于临床研究与应用. A reversed - phase HPLC method has been established for the analysis of valproic acid in human serum, using 4 - bromomethyl - 7 - methoxycoumain as derivative reagent which transformed valproic acid into a ester detectable with UV - detector. The ratio of peak heights correlated linearly with concentration of valproic acid ranging from 15 to 200μg/ml, r = 0. 9991. The mean recovery is 101. 8% . The method is sensitive, accurate, selective, simple and fast. The method has been applied in the therapeutic drug monitoring of 34 patients, in which acquired satisfied effects, showing that the method is suitable for the clinical research and application.
