高产Monacolin K红曲霉菌种的诱变选育及液态发酵工艺优化 被引量:12

Mutagenic breeding of the high-yield Monacolin k strains of Monascus sp. and process optimization its submerged fermentation

摘要 从29株红曲霉菌(Monascus sp.)中筛选得到的M2为出发菌株,进行UV照射、LiCl诱变以及硫酸二乙酯处理与LiCl复合诱变三次诱变,筛选获得Monacolin K的产量显著提高的菌株MS-12。对液态发酵培养基、发酵温度、接种量以及摇床转速等因素进行优化。在最优条件下MS-12液态发酵Monacolin K的产量显著提高,达到264.7μg/mL,桔霉素含量仅为0.026ng/mL。 From 29 strains of Monascus sp. ,one strain(M2) which produced Monacolin K higher than others was chosen to be original strain. The strain then treated with UV,LiCI,ethyl sulfate and LiCI,a mutant strain of Monascus sp. named MS-12 was obtained which could produce much higher Monacolin K than M2. These optimizations of the liquid-state fermentation medium,fermentation temperature,inoculum concentration and the shaking speed were done. Under optimal conditions,the Monacolin K content of MS-12 improved significantly, reached 264.7ug/mL. And the citrinin content was only 0.026ng/mL.
