耐力训练对急性酒精性肝损伤大鼠线粒体融合与分裂的影响 被引量:1

Effect of Endurance Training on the Mitochondrial Fusion and Fission Dynamic in Alcohol-Induced Acute Hepatic Injury

摘要 目的:研究预训练对急性酒精性肝损伤大鼠肝脏线粒体融合/分裂的变化。方法:以SD大鼠建立急性酒精性肝损伤模型,以无负重游泳为运动手段,测定肝脏融合蛋白Mfn1/2和Opa1与分裂蛋白Drp1基因的表达以及肝脏H2O2的含量。结果:急性酒精摄入导致肝脏融合蛋白Mfn1/2和Opa1基因表达显著降低,而分裂蛋白Drp1显著升高,肝脏H2O2含量显著升高;耐力训练后肝脏融合蛋白Mfn1/2和Opa1基因表达显著升高,而分裂蛋白Drp1显著降低,肝脏H2O2含量显著降低;耐力训练后,再给予急性酒精摄入,肝脏线粒体融合蛋白Mfn1/2和Opa1基因表达恢复到正常水平,而分裂蛋白Drp1和肝脏H2O2含量依然显著高于对照组。结论:预训练可以有效减少急性酒精摄入引起的肝脏活性氧的产生,减弱线粒体分裂,增强线粒体融合。 Objective:This study aims to investigate the influence of pre-endurance training on mitochondrial fusion and fission in alcohol-induced acute hepatic injury rats. Methods: Measured the expressions of Mfnl/2, Opal and Drpl mRNA, and the content of H2O2 in liver tissue in alcohol-induced acute hepatic injury rats after 12 week un- load swimming training. Results: Compared with the control group, the mRNA expressions of Mfnl/2 and Opal decreased, the expression of protein Drpl increased significantly, and the content of H202 in liver tissue signifi- cantly increased in acute alcohol intake rats. After 12 week endurance training, the mRNA expressions of Mfnl/2 and Opal increased, Drpl protein expression decreased significantly, and the content of H2O2 in liver tissue signif- icantly decreased. Then after acute alcohol intake, the mRNA expressions of Mfnl/2 and Opal recovered to nor- mal level, while Drpl protein expression and the content of H2O2 still significantly were higher than the control group. Conclusion: Pre-endurance training can decrease acute alcohol administration-induced reactive oxygen spe- cies (ROS) formation which can decrease mitochondrial fission and increases mitochondrial fusion.
