摘要
目的 :测定、比较经腹腔动脉和周围静脉注射 5 -氟尿嘧啶 (5 -Fu)后体循环周围静脉血及胰腺、肝脏组织内的药代动力学变化。方法 :家兔 2 0只 ,随机分为腹腔动脉给药组 (CA) (n =1 0 )和周围静脉给药组 (PV) (n =1 0 ) ,按 40mg/kg体重注入 5 -Fu。注药后定时取颈动脉血及胰腺和肝脏组织。高效液相色谱法 (HPLC)测定血及组织中的 5 -Fu浓度。结果 :CA组和PV组快速注射大剂量的 5 -Fu后 ,血中的药物浓度变化相似。CA给药能提高胰腺的药物浓度 ,在给药后 1 0、2 0、30分钟CA组是PV组的 57、2 9、6倍。 60分钟时两者相似。 60分钟后PV组又以较CA组为快的速度衰减。CA给药和PV给药肝脏的药物浓度变化和胰腺相似。结论 :CA给药可在 60分钟内提高胰腺、肝脏的药物浓度 ,从而提高胰腺癌化疗效果并预防肝转移的发生。
Objective:To measure the fluorouracil (5-Fu) content in blood of the pancreas and liver of rabbits after injection from coeliac artery (CA) and peripheral vein (PV) respectively.Methods:Twenty rabbits were divided into CA group (n=10) and PV group (n=10),40 mg/kg 5-Fu was given by CA or PV quickly.Drug level in the common carotid artery and the pancreas and the liver were measured at different time intervals,by the high performance liquid clizomatographic (HPLC).Results:There was no marked difference in the plasma 5-Fu level and blood concentration-time curves following CA or PV injection of 5-Fu.However,the distribution of 5-Fu in the pancreas given by CA was quite different from those of PV.At 10、20、30 minutes after CA injection,the 5-Fu content in the pancreas was showa to be 57,29 and 6 fold high than those of PV injection respectively,the 5-Fu content in the liver was showu to be 38、22 and 3 7 fold high than those of PV injection respectively.Conclusions:Intra CA chemotherapy of 5-Fu can maintain a stable and persistent high drug level in the pancreas and the liver.Those pharmacokinetic properties of 5-Fu are of advantage in preventing and treatiug recurrence of pancreas carcinoma and metastasis in the liver.
