喉鳞癌组织中PTTG的表达及与bFGF和微血管密度的关系

Expression of PTTG in laryngeal squamous cell carcinoma and its relationship with bFGF expression and MVD

摘要 目的探讨喉鳞癌组织中垂体肿瘤转化基因(PTTG)和碱性成纤维细胞生长因子(bFGF)的表达以及在新生血管生成过程中的作用。方法应用免疫组化SP法分别检测58例喉鳞癌组织和30例癌旁组织中PTTG和bFGF两种蛋白的表达情况,并用CD34标记并计数微血管密度(MVD)。结果 PTTG和bFGF在喉鳞癌组织中的表达分别为72.41%和84.48%,均高于癌旁组织的33.33%和36.67%(P<0.05);bFGF和PTTG的表达与喉鳞癌淋巴结转移、分化程度及临床分期均密切相关(P<0.05),且两者在喉鳞癌组织中表达呈显著正相关(P<0.05)。MVD值在喉鳞癌组织中表达明显大于喉鳞癌旁黏膜(P<0.05),并与喉鳞癌淋巴结转移呈显著相关(P<0.05)。MVD分别与喉鳞癌组织中PTTG和bFGF表达呈正相关(P<0.05)。结论 PTTG和bFGF在喉鳞癌组织中的高表达与MVD关系密切,在喉鳞癌发展中可能起重要作用。两者联合检测有助于喉鳞癌的诊断和恶性程度评估。 Objective To investigate the expressions of pituitary tumor transforming gene (PTTG) and basic fibroblast growth factor(bFGF) in laryngeal squamous cell carcinoma(LSCC) and the role of them in the angiogenesis. Methods Immunohistochemistry was used to detect the expressions of PTTG, bFGF and microvessels density(MVD) in 58 samples of LSCC(group A) and 30 samples of carcinoma-adjacent normal tissues(group B). Results The expressions of bFGF and PTTG in group A were 72.41% and 84.48 %, respectively, which were significantly higher than 33.33% and 86.67% in group B(P〈0. 05). The expressions of bFGF and PTTG were closely associated with tumor differentiation, lymph node metastasis and clinical stage (P〈0. 05). A signifcant positive correlation was found between bFGF and PTrG expressions(P〈0. 05). The value of MVD in group A was significantly higher than that in group B(P〈0. 05), which was positively correlated with lymph node metastasis(P〈0. 05). Conclusion The overexpressions of PTTG and bFGF are closely related with MVD in LSCC,which might play an important role in the development of laryngeal carcinoma. Combined detection of PTTG and bFGF may be helpful in the diagnosis and evaluation of the severity of laryngeal carcinoma.
