【摘要】目的观察大鼠骨骼肌缺血再灌注过程中肝脏功能的变化及外源性H。S吸人对其产生的影响。方法雄性Wistar大鼠69只,应用结扎止血带法复制大鼠双下肢缺血再灌注模型,随机分为10组。①c组(正常对照组,n=8);②12组(缺血2h组,n=4);③14组(缺血4h组,n=8);@IR0.5组(缺血再灌注0.5h组,n=8);⑤IR2组(缺血再灌注2h组,n=8);@IR4组(缺血再灌注4h组,n=8);(7)IR6组(缺血再灌注6h组,n=8);(8)IR12组(缺血再灌注12h组,n=4);(9)IR24组(缺血再灌注24h组,n=6);⑩H2S吸入组(大鼠肢体缺血4h后,再灌注时给予含80ppmH2S的合成空气持续吸入4hn=7)。观察各组大鼠血浆GPT、GOT、H2S水平的变化。结果与正常对照组比较,缺血4h组血浆GPT、GOT水平开始上升,P〈0.05,再灌注后继续上升并于4—6h达峰值,明显高于缺血4h组P〈0.05。给予外源性H2S吸入后血浆H2S含量明显升高,P〈0.05,同时血浆GPT、GOT含量明显降低,P〈O.05。结论骨骼肌缺血再灌注可导致肝功能损伤,外源性H2S吸入对其具有保护作用。
Objective To explore the changes of liver function after skeletal muscle ischemia reperfusion and observe the effect of exogenous hydrogen sulfide (H2S) on the liver function after hindlimb ischemia reperfusion. Methods IR models were established with 69 male Wistar rats by strapping the both hind limbs with tourniquet. The rats were randomly divided into 10 groups as (C) group normal, (I2) group 2h-ischemia, (I4) group 4h-ischemia, (IR0.5) groups 4h-ischemia with 0.5 h, (IR2) 2 h, (IR4) 4 h, (IR6) 6 h, (IR12) 12 h and (IR24) 24 h reperfusion, (H^S) Group inhaling 80 ppm H2S for 4 h after ischemia reperfusion 4 h. The plasma levels of GPT, GOT, H2S were measured. Results Compared With group normal, the GPT, GOT levels in plasma were remarkably increased in group I4 ,P 〈 0.05, while the levels further increased in the reperfusion groups, and reached peak value in group IR4, IR6. The plasma H2S level was decreased obviously in group ischemia reperfusion ,P 〈 0.05. The plasma H2S level was significantly higher, and the plasma levels of GPT and GOT were markedly decreased after inhaling 80 ppm H2S ,P 〈 0.05. Conclusions Skeletal muscle ischemia reperfusion is considered to lead liver function damage, and exogenous H2S inhalation may protect liver function from skeletal muscle ischemia-repefusion injury.
