外源性H2S对大鼠肢体缺血再灌注后肾损伤的保护作用 被引量:1

摘要 目的研究外源性H2S吸人在保护大鼠肢体缺血再灌注后肾损伤中的作用。方法雄性Wistar大鼠69只,随机分为10组。①正常对照组(c组,n=8);②缺血2h组(12组,n=4);③缺血4h组(14组,n=8);④缺血再灌注0.5h组(IR0.5组,n=8);⑤缺血再灌注2h组(IR2组,n=8);⑥缺血再灌注4h组IR4组(,n=8);⑦缺血再灌注6h组(IR6组,n=8);⑧缺血再灌注12h组(IR12组,n=4);⑨缺血再灌注24h组(IR24组,n=6);⑩H2S吸入组(H2S组,n=7)。观察各组大鼠血浆Cr、BUN及血浆H2S的变化。结果与正常对照组相比较,缺血再灌注4h组血浆Cr、BUN含量均明显升高(P〈0.05),血浆H2S含量明显下降(P〈0.05)。给予外源性H2S吸入后血浆Cr含量明显降低(P〈0.05),同时血浆H2S含量明显升高(P〈0.05)。结论外源性H2S吸入对骨骼肌缺血再灌注致肾功能损伤发挥保护作用。 Objective To observe the safety of exogenous hydrogen sulfide (H2S) on the kidney injury after rat hindlimb isehemia reperfusion. Methods 69 Wistar rats were randomly divided into ten group: 1. Control group (n=8), 2. Group of 2-hour ischemia (I-2, n=4), 3. Group of 4-hour ischemia(I-4, n=8), 4. Group of 4-hour ischemia with 0.5-hour (IR0.5, n=8), 5. 2-hour (IR2, n=8), 6. 4-hour (IR4, n=8), 7. 6-hour (IR6, n=8), 8. 12-hour (IR12, n=4), 9. 24-hour (IR24, n=6), 10. Group H2S (H2S, n=7). The plasma levels of Cr, BUN and H2S of each group were measured. Results Comparing with the control group, the plasma Cr, BUN level significantly increased in group ischemia reperfusion (P 〈0.05), the plasma H2S level was decreased obviously in group ischemia reperfusion (P 〈0.05). The plasma H2S level was significantly higher and the plasma levels of Cr were markedly decreased after inhaling 80 ppm H2S (P 〈0.05). Conclusion Exogenous H2S inhalation is considered being effective in protecting kidney function from muscle ischemia-repefusion injury.
