摘要
目的 :观察丹皮酚 (Pae)对人胎儿主动脉平滑肌细胞增殖的抑制作用。方法 :利用体外细胞培养技术 ,用人高脂血清造成平滑肌细胞 (SMC)增殖的模型并通过 MTT测定、细胞计数及透射电镜下的细胞超微结构的观察等方法 ,观察了 Pae对人胎儿平滑肌细胞的保护作用。结果 :人高脂血清可明显刺激人胎儿平滑肌细胞的增殖并引起其表型的改变 ,而 Pae可呈浓度依赖性地抑制平滑肌细胞的增殖并可显著抑制高脂血清对 SMC的促增殖作用。结论 :Pae抗动脉粥样硬化的作用可能部分是由于抑制 SMC异常增殖的结果。
objective:To study the effect of paeonol(Pae) on increased proliferation of cultured aortic smooth muscle cells(SMC)from human by hyperlipid serum.Methods:In this study,cell cul ture technique in vitro was used to investigate the protective effect of paeonol on the cultured aortic smooth muscle cells from human by measuring MTT metablism and cell numbers and observing cell ultrastructure under TEM.Results:The results showed that the hyperlipid serum caused the proliferation of SMC.The Pae can inhibit the proliferation of SMC dose-dependently and can inhibit the proliferation of SMC stimulated by hyperlipid serum significantly.Conclusion:the Pae may prevent atherosclerosis and inhibit progression of the atheroslerotic lesions by interfering with the proliferation of arterial SMC.
