摘要
目的 :建立无机有毒粉尘颗粒致 AMs产生 H2 O2 的电镜原位显示技术。方法 :以铈离子为捕捉剂 ,于产生 H2 O2 的原位形成过氢氧化铈沉淀。结果 :H2 O2 的阳性反应呈线状高电子密度沉淀 ,分布于 AMs质膜、吞噬体 /吞噬泡膜、及溶酶体膜。结论 :无机有毒粉尘颗粒致 AMs产生的 H2 O2 能够以电镜细胞化学方法于原位显示。该技术在显示氧自由基 H2 O2 的同时 ,能同步观察细胞的超微结构。因而比自由基总体测定技术更能直观具体地体现 AMs与粉尘相作用的关系 ,是一项实用的实验室技术。
To set up a morphological technique to demonstrate the hydrogen peroxide(H 2O 2) in situ on alveolar macrophages (AMs) introduced by mineral dusts. Methods: Using cerium anion as a captor, to product cerium perhydroxide precipitate in si tu of the production for H 2O 2. Results:The positive reaction of H 2O 2 w as expressed a high electronic dense deposits by the electron-opaque cerium per h ydroxide, exhibiting a linear profile. The precipitation of H 2O 2 was localiz ed on the plasma membrane, the membrane of the phagosome and/or pinosome, and th e lysosomal membrane. Conclusion:It was done to demonstrate H 2O 2 in situ o n AMs introduced by mineral dusts. This technique is possible to observe the rea ction of H 2O 2 and to observe the cellular ultrastructure at the same time. I n situ evaluation of H 2O 2 oxygen free radical is superior to the other techn iques in a sense. It is more convenient to directly study the real situation of the relationship between the production of oxygen free radical and the cell dama ge introduced by mineral dusts. It affords a useful experimental research tec hnique.
