摘要
目的观察从肝治心组方(CGZXP)对冠心病心肌缺血大鼠局部缺血心肌血管生成素-1(Ang-1)、血管生成素-2(Ang-2)蛋白表达,以及对缺血血管新生作用的影响。方法 SD大鼠按随机方法分成假手术组、模型组、麝香保心丸组和从肝治心方组4组。采用冠脉结扎法诱导急性心肌梗死模型,造模24 h后给药,每日1次。于造模14 d后处死大鼠,取出心脏,肉眼观察心肌情况,应用免疫组织化学法进行缺血心肌Ang-1、Ang-2蛋白表达的检测。结果从肝治心方组对改善大鼠活动、对外界反应、精神、食欲、皮毛质感等均优于其他各组;免疫组化结果提示从肝治心方组和麝香保心丸组的心肌细胞Ang-1的表达高于模型组,Ang-2的表达低于模型组(P均<0.01),但两组之间比较差异无统计学意义。结论 CGZXP通过促进Ang-1、抑制Ang-2的表达,可能促进血管新生,从而达到增加心脏侧支循环,抗心肌缺血的作用。
Objective: To observe the effects of Conggan Zhixin Prescription(CGZXP) on myocardial ischemic of Ang-1 and Ang-2 protein expression and to promote the role of ischemic angiogenesis on myocardial ischemic in rats. Methods: According to completely random method SD rats were divided into 4 groups: Sham operation group,the model group,the Shexiangbaoxin pill group and Conggan Zhixin Prescription group. Acute myocardial infarction was induced by coronary artery ligation model. Acute myocardial infarction animal models were induced by ligating the coronary artery, after model rats were made for 24 hours, giving them medicine 1 times a day.Rats were killed 14 days after modeling, remove the heart, observe cardiac conditions by naked-eyes. Angiogenesis and Ang-l, Ang-2 protein expression of ischemic myocardial were determinated and detected immunohistochemistry. Results: The basic situation of Conggan Zhixin Prescription group including rats' activities,respond to the outside world, spirit, appetite ,fur, texture, etc was better than the other groups ;The results of alkaline phosphatase staining suggested that Conggan Zhixin Prescription group and Shexiangbaoxin pill group may reduce the extent of my- ocardial ischemia;Immunohistochemical results suggested that the expression of Ang-1 cardiomyocytes Conggan Zhixin Prescription group and Shexiangbaoxin pill group is higher than the model group ,Ang-2 expression is low- er than the model group, It is not statistically significant between the two groups. Conclusion: CGZXP can effec- tively promote neovascularization in ischemic myocardium, thereby reducing the infarct size and improving cardiac function;CGZXP promote the expression of Ang-1 and inhibit Ang-2 to promote angiogenesis,so as to achieve the increased cardiac collateral circulation and improve ischemic.
