HPLC法测定新疆白喉乌头不同部位氢溴酸高乌甲素的含量

摘要 目的比较新疆尼勒克地区白喉乌头不同部位(根、茎、叶)中氢溴酸高乌甲素的含量。方法采用X Bridge C18生物碱专用色谱柱(250mm×4.6mm,5μm),以0.04mol/L醋酸铵-乙腈(63∶37)为流动相等度洗脱,流速0.8mL/min,柱温40℃,检测波长为252nm,进样量10μL高效液相色谱法测定白喉乌头根、茎、叶中氢溴酸高乌甲素含量。结果白喉乌头中氢溴酸高乌甲素在根中含量为0.203 8mg/g,在叶中的含量为0.061 1mg/g,在茎中的含量为0.011 1mg/g。结论白喉乌头根中氢溴酸高乌甲素含量高于叶和茎中,为白喉乌头资源的进一步利用奠定了一定基础。 Objective The study is to compare hydrobromide lappaconitine content in different parts(root, stem, leaf) ofAconitum leucostomurn Worosch by using HPLC method. Methods Use X Bridge C18 Alka- loid chromatographic column (250 mmX4.6 mm,5 μm) and take 0.04 mol/L solution of ammonium ace- tate- acetonitrile as solvent to elute with same flow rate of 0. 8ml/min under the column temperature 40℃. The detection wavelength is set as 252 nm, and injection volume was 10 μL. Results The content of hydrobromide lappaeonition in the root ofAconitum leucostornurn Worosch is 0. 203 8 mg/g, 0. 061 1 mg/g in leaf and 0. 011 1 mg/g in stem. Conclusion The content of hydrobromide lappaconition in Aconitum leucostornum Worosch in root is higher than that is in leaf and stem. The studies lay the foundation for fur- ther application of Aconitum leucostornum Worosch resources.
