白喉乌头挥发油的GC-MS分析 被引量:5

摘要 目的:分析鉴定白喉乌头挥发油的组成组分,为其进一步的开发利用奠定基础。方法:取白喉乌头干粉250g,加蒸馏水1L,用电热套加热回流制备其挥发油,然后对挥发油直接进行GC-MS分析。色谱条件:RTX-5MS石英毛细管色谱柱(30m×0.32mm×1.0μm);升温程序:60℃保持1min,以10℃.min-1升至300℃,保持5min;载气(99.999%He),流速1.0mL.min-1,压力3.4kPa,进样量1μL;分流比30∶1。质谱条件:电子轰击(EI)离子源,电子能量70eV,传输线温度250℃,离子源温度200℃,质量扫描范围m/z30~500。采用Nist147谱库及Wiley7谱库进行检索。结果:分离出105个化合物,鉴定了其中的72个化合物,已鉴定出的化合物占总挥发油质量的86.81%。其中含量最高的成分为顺,顺-亚油酸(占29.48%)和n-棕榈酸(占21.08%);含量较高的成分有芳姜黄酮(4.78%)、十五烷酸(3.3%)、1,4-顺-1,7-反-菖蒲烯酮(2.85%)、2-羟基环十五烷酮(2.83%)、亚油酸甲酯(2.5%)、亚油酸乙酯(1.55%)和棕榈酸甲酯(1.41%)。结论:分析结果表明,白喉乌头挥发油中亚油酸和棕榈酸含量比较高,二者含量超过50%,具有一定的开发应用价值。 Objective:To analyze and identify the ingredients of volatile oil from Aconitum leucostomum Worosch.for its further exploitative use. Methods:250 g of Aconitum leucostomum dry powder and 1 L distilled water were mixed,and the volatile oil was extracted by refluxing through electric heating jacket,and then analyzed by GC-MS.Chromatographic conditions:RTX-5MS quartz capillary column(30 m×0.32 mm×1.0 μm);temperature program:maintain in 60 ℃ about 1 min,elevated by 10 ℃·min-1 to 300 ℃,remain 5 min;carrier gas(99.999% He),velocity 1.0 mL·min-1,pressure 3.4 kPa,the injection volume 1 μL;split ratio 30:1.MS conditions:electron impact(EI)ion source,electron energy 70 eV,transmission line temperature 250 ℃,ion source temperature 200 ℃,mass scan range m/z 30-500.And the resultes were contrasted with Nist 147 spectral library and Wiley 7spectral library. Results:105 compounds were separated,and 72 compounds were identified from the volatile oil and cis,cis-linoleic acid(accounts for 29.48%),n-hexadecanoic acid(accounts for 21.08%)are major ingredients.Furthermore,the ingredients with more contents are ar-turmerone(4.78%),pentadecanoic acid(3.3%),1,4-cis-1,7-trans-acorenone(2.85%),2-hydroxy-cyclopentadecanone(2.83%),methyl linoleate(2.5%),linoleic acid ethyl ester(1.55%),methyl palmitate(1.41%).Conclusion:Linoleic acid and hexadecanoic acid have higher contents and overall contents of both of them are more than 50%,and they have certain exploitive and practical value.
