摘要
目的探讨X染色体连锁凋亡抑制蛋白相关因子1(XAF1)基因在肺挫伤后细胞凋亡中的作用。方法 25只新西兰白兔随机均分为五组:肺挫伤后6h组(A1组)、24h组(A2组)、48h组(A3组)、72h组(A4组)及正常对照组(C组);A1、A2、A3和A4组采用自由落体撞击装置建立兔肺挫伤模型。TUNEL法检测肺挫伤后细胞凋亡,RT-PCR和免疫组化方法检测XAF1mRNA及蛋白在肺组织中的表达。结果与C组相比,A1、A2、A3、A4组肺组织细胞凋亡显著增多,XAF1mRNA及其蛋白表达明显升高,48h达峰值(P<0.05)。结论肺挫伤诱导XAF1mRNA和蛋白表达增加,调控细胞凋亡。
Objective To investigate the effect of X-linked inhibitor of apoptosis (XIAP)- associated factor I(XAF1) in cell apoptosis following pulmonary contusion(PC). Methods Twenty- five rabbits were randomly and equally divided into five groups of A1 (6 h after PC), A2 (24 h after PC),A3(48 h after PC), A4(72 h after PC) and C(normal control). Free fall hitting was used to establish the rabbits PC models. The lung specimens were taken at 6 h(group A1), 24 h(group A2), 48 h(group A3),72 h(group A4) and from group C. The expressions of XAF1 mRNA and protein were detected by RT-PCR and immunohistochemistry, and cell apoptosis was examined by TUNEL method. Results Compared with group C, the lung cell apoptosis was increased and the XAF1 gene and protein expressions were higher(P〈0. 05), which peaked at 48 hours after PC in groups of A1, A2,A3 and A4. Conclusion Pulmonary contusion induces the expressions of XAF1 mRNA and protein,and regulates cell apoptosis in the progression of lung injury after lung contusion.
