摘要
目的:探讨bcl-2、bax在胸部撞击致大鼠急性肺损伤(ALI)中的表达改变及其意义。方法:健康雄性SD大鼠24只,随机分为正常对照组和创伤后3h,12h,24h组。采用TUNEL法以及免疫组化技术观察肺组织bcl-2、bax表达及细胞凋亡的变化,观察病理形态。结果:与对照组比较,各创伤组肺组织内细胞凋亡指数增加,并随着病程延长逐渐上升(P<0.05);bax基因表达明显增强,并随着病程延长而逐渐增高(P<0.01);bcl-2基因表达亦明显升高,但随着病程延长呈降低趋势(P<0.01);bcl-2/bax比值降低,该比值也有随着病程延长而逐渐下降的趋势(P<0.01)。各创伤组光镜下见肺组织损伤明显。结论:肺组织细胞凋亡及bcl-2、bax基因表达参与胸部撞击致ALI时的肺损伤,并在其发病机制中起着一定的作用。
Objective:To evaluate the effects of apoptosis and study the expression ofapoptosis related genes hcl-2 and bax in acute lung injury following blunt chest trauma in rats.Methods:An animal model of acute lung injury was established in SD rat by impacting chest.24 SD rats were divided equally and randomly into normal group (n=6).ALI model group (include 3h group,12h group,24h group;n=6 eaeh).The pathological changes and cell apoptosis were observed with TUNEL,immunochemieal methods.Results:As compared with M group,in every group after chest trauma.apoptotic index markedly increased,and increased gradually with the course advanced (P〈0.05);expressions of bax and bcl-2 also markedly increased,and expressions of bax increased gradually with the course advanced,but expressions of bcl-2 was reduced gradually with the disease prolonged (P〈0.01);the ratio of bcl-2/bax markedly decreased, and reduced gradually with the disease prolonged (P〈0.01);lung tissue injuries were significant under a lightmicroscope.Conelusion:The apoptosis and expression of bax and bcl-2 in lung tissue might be involved in pathogenesis of the lung injury lbllowing blunt chest trauma
