凝血酶敏感蛋白-1对体外培养的大鼠肺成纤维细胞的影响

摘要 目的了解凝血酶敏感蛋白-1(TSP-1)对体外培养的大鼠肺成纤维细胞(LF)的影响。方法体外培养大鼠LF,加入不同浓度(10-12、10-11和10-10mol/L)TSP-1处理12、24和48 h,采用四唑盐(MTT)法检测细胞增殖,流式细胞术检测细胞周期。结果 TSP-1刺激后,MMT法实验显示TSP-1显著促进大鼠LF增殖,此作用呈浓度和时间依赖性。FCM分析显示TSP-1处理LF 12 h后,G1期比例下降,G2+S期细胞比例即出现增加,48 h达高峰。结论 TSP-1呈浓度和时间依赖方式促进大鼠LF增殖。
