摘要
目的探讨碘化钾(KI)对人皮肤成纤维细胞(HSF)胶原合成的影响。方法将体外培养的HSF分为5组,KI组分别加入3、30、300 mg/L的KI,bFGF组加入3 mg/L bFGF,对照组加入同等剂量的无菌生理盐水,作用96 h,倒置显微镜下观察各组HSF形态学变化,采用Real-Time PCR法检测Ⅰ型胶原(COL1)、Ⅲ型胶原(COL3)mRNA,采用3H-脯氨酸比色法测定成纤维细胞胶原的合成。结果与对照组比较,3 mg/L KI组COL3 mRNA表达升高,bFGF组COL1、COL3 mRNA表达升高(P均<0.05);与对照组比较,3、30 mg/L KI组及bFGF组培养液中胶原合成增高,3、30、300 mg/L KI组及bFGF组细胞内胶原合成增高(P均<0.05)。结论适量浓度的KI可使人皮肤成纤维细胞合成和分泌胶原增加。
Objective To investigate the effects of KI on the collagen synthesis of skin fibroblasts in vitro.Methods Human skin fibroblasts (HSF) cultured in vitro were divided into 5 groups:the control group (treated with the same dose of sterilizing saline),3 mg/L KI group,30 mg/L KI group,300 mg/L KI group and 3 mg/L bFGF group.After being stimulated by different concentrations of KI for 96 hours,morphological changes of HSF were observed by inverted microscope,type Ⅰ collagen (COL1) and type Ⅲ collagen (COL3) mRNA was detected by real-time PCR and collagen synthesis of HSF was examined by hydroxyproline colorimetric analysis.Results Compared with the control group,COL3 mRNA expression in 3 mg/L KI group was significantly increased,COL1,COL3 mRNA expression was also increased in bFGF group (all P < 0.05).The collagen synthesis in culture solution of 3mg/L KI group,30mg/L KI group and bFGF group was remarkably increased; the collagen synthesis in the supernatant of cell of 3 mg/L KI group,30 mg/L KI group,300 mg/L KI group and bFGF group was also increased (all P < 0.05).Conclusion Proper concentrations of KI promote cell synthesis of HSF and secretion of collagen.
