Detection of Organic Acids in Grape and in Grape Wine by HPLC with Double Columns

摘要 探索优化利用双小柱检测葡萄及葡萄酒中8种主要有机酸的方法,并用此方法检测山西乡宁地区葡萄及葡萄酒中8种主要有机酸。结果表明,以20℃、210 nm、pH2.1的磷酸氢二钾和硫酸铵溶液作为流动相,可以将8种主要的有机酸完全分离检出,且回收率高、重复性好。葡萄中可检出5种有机酸,其中,以酒石酸和L-苹果酸的含量最高,且随成熟度的增加含量降低;葡萄酒中可以检出7种有机酸,主要是酒石酸、乳酸、乙酸。利用此方法可以监控葡萄的成熟度以及葡萄酒的发酵过程,提高葡萄酒的品质。 HPLC with double columns was used to detect organic acids in grape and in grape wine. In the experiment, organic acids in grape and in grape wine from Xiangning county, Shanxi province were detected. In the conditions of temperature at 20 ~C, 210 nm, pH 2.1 and dipotassium hydrogen phosphate and ammonium sulfate solution used as mobile phase, 8 major organic acids could be completely separated with good recov- ery and reproducibility. 5 organic acids were detected in grape, the content of tartaric acid and malic acid was the highest, and their content de- creased with the the enhancement of grape maturity. 7 organic acids were detected in grape wine, and they were mainly tartaric acid, lactic acid and acetic acid. The application of such method was convenient for monitoring grape maturity and controlling the fermentation of grape wine to improve grape wine quality.
