摘要
目的:比较周麦16、Spelt7、中优9507、Spelt53、小偃54的小麦胚芽胚乳中蛋白质、赖氨酸和谷胱甘肽含量和性质差异。方法:蛋白质性质分析采用SDS-聚丙烯酰胺凝胶电泳(SDS-PAGE)和聚丙烯酰胺等电聚焦电泳(IEF-PAGE)方法;蛋白质、赖氨酸和谷胱甘肽含量测定分别采用Folin-酚法、茚三酮法和DTNB法。结果:同一品种胚芽、胚乳间蛋白质亚基组成及数目有所不同;品种间胚芽、胚乳中蛋白质亚基分子质量和等电点分布范围相似;品种间胚芽蛋白质含量及谷胱甘肽含量普遍存在显著差异(P<0.05)或极显著差异(P<0.01);同一品种内胚芽蛋白质、赖氨酸和谷胱甘肽含量通常高于胚乳,且差异显著(P<0.05)或极显著(P<0.01)。结论:小麦胚芽蛋白质和谷胱甘肽普遍存在种质差异,且三者含量胚芽普遍高于胚乳。总体评价以中优9507品质最好。
Purpose: To compare the content and properties of protein,lysine and glutathione extracted from wheat germ or endosperm of five cultivars (Zhoumai 16, spelt 7, Zhongyou 9507, spelt 53 and Xiaoyan 54). Methods: Protein properties were characterized by SDS-PAGE and IEF-PAGE. The contents of protein, lysine and glutathione were quantified and evaluated using Folin-phenol assaying, ninhydrin reaction and DNTB circular reaction, respectively. Results: The compositions and numbers of protein subunits of wheat germ were different from that of endosperm in each cultivar. The molecular weights and isoelectric points of protein subunits had both similar distribution range between wheat germ and endosperm in five cultivars. The contents of wheat germ protein and glutathione generally were distinct among different cultivars with significant difference (P﹤0.05 or P﹤0.01). Protein, lysine and glutathione of wheat germ of the same cultivar usually were much more than that of endosperm with significant difference (P﹤0.05 or P﹤0.01). Conclusions: Wheat germ protein and glutathione currently existed diversities resulting from germplasm difference, and the contents of protein, lysine and glutathione of wheat germ were commonly higher than that of endosperm. As a whole, Zhongyou 9507 was the best in quality.
