一种吖啶-4-酰胺类探针的制备及其与牛血清蛋白作用的研究 被引量:1

摘要 制备一种吖啶-4-酰胺类探针9-[(N-2-二甲氨乙基)吖啶-4-酰胺]-α-丙氨酸(NACA),研究NACA与牛血清蛋白(BSA)的结合作用.测定了Stern-Volmer猝灭常数KSV、热力学参数(ΔH、ΔG及ΔS)、结合位点数n、表观结合常数KA和NACA-BSA作用距离r,确定了NACA对BSA的荧光静态猝灭机制和主要作用力. An acridine - 4 - carboxamide skeleton - based probe N - (9 - ( N - ( 2 - dimethylamino) ethyl) acridine - 4 - carboxamide) - α - alanine (NACA) was synthesized. The interaction between NACA and bovine serum albumin (BSA) was investigated. The Stern - Volmer fluorescence quenching constant (Ksv) was investigated, as well as thermodynamics constants of △H, △G and △S. Other param- eters including the number of combining sites ( n), apparent combining constants ( KA ) and interaction distance (r) of NACA -BSA were estimated. The static quenching was found to cause the fluorescence quenching of BSA and the interaction mechanism between NACA and BSA was also discussed.
