摘要
目的:观察不同剂量痰热清对急性肺损伤大鼠MMP-9的影响。方法:雄性Wistar大鼠72只,随机分为4组,分别是正常对照组(A组)、急性肺损伤模型组(B组)、常规剂量组(C组)、高剂量组(D组),每组18只。后3组大鼠经尾静脉静注脂多糖(LPS)5 mg/kg制成ALI模型。30 min后经腹腔分别注射生理盐水、生理盐水、痰热清(2.1 mL/kg)、痰热清(4.2 mL/kg)。每天早上给药1次,连续给药3 d。每次给药后6 h将每一组的6只大鼠经股动脉放血处死,取肺组织测量肺湿/干重比,检测肺匀浆的MMP-9含量,并在光镜下观察肺组织形态学变化。结果:C、D两组与B组相比,W/D和肺匀浆MMP-9含量有不同程度的下降,具有显著性差异(P<0.05或P<0.01);而C、D两组之间则无显著性的差异(P>0.05),而D组大鼠出现不良反应和死亡的情况较C组大鼠多。光镜下B组呈现急性肺损伤的病理改变,而C、D两组较B组有明显改善,两组间无明显差异。结论:痰热清可以降低ALI大鼠肺W/D和肺匀浆的MMP-9含量,常规剂量和高剂量效果相当,但高剂量可能会导致不良反应增加。
Objective : To evaluate the effects of different doses of Tanreqing Injection on MMP - 9 of the ALI rats. Method:72 Wistar male rats were randomly divided into 4 groups, 18 in per group. They were normal control group (group A) ,ALI model group (group B) ,normal dose group (group fi) and high dose group (group D). The rat model of ALl was produced with intravenous injecting lipopolysaccharide. After 30 minutes, we respectively gave them normal sodium, normal sodium,Tanreqing (2.1 mL/kg) ,and Tanreqing (4.2 mL/kg) ,for 3 days consecutively. 6 hours after each ad- ministration, the content of MMP -9 in the rats' lung homogenate, the wet weight -dry weight ratio(W/D) and patholog- ic change of lung were compared. Result : The indexes of MMP - 9 and W/D of group C and D were lower than those of group B had significant difference to some extent( P 〈 0.05 or P 〈 0.01 ) , while no significant difference could be found between group C and D( P 〉0.05 ). But incidence of adverse effect and the mortality of group D seemed to be higher than that of group C. The ALI pathologic change could he found in group B with microscope, while group C and D improved significantly, but no significant difference between them. Conclusion:Tanreqing Injection can reduce W/D and the content of MMP -9 in the rats' lung homogenate, and normal dose has similar effect to the high dose, but high dose seems to in- crease the incidence of adverse effect.
