不同剂型补阳还五汤对窒息性大鼠心肺复苏后NSE水平的干预作用 被引量:1

Intervention of Different Formulations of Buyang Huanwu Decoction in NSE Level in Asphyxiating Rats Post Cardiopulmonary Resuscitation

摘要 目的:采用大鼠窒息型心脏骤停模型,观察血清和脑组织匀浆液神经元特异性烯醇化酶(NSE)浓度,并用不同剂型补阳还五汤剂进行干预,研究大鼠心肺复苏(CPR)后血清和脑组织匀浆NSE浓度的影响。方法:成年雄性Sprague-Dauley(SD)大鼠32只,随机分为假手术对照组(A组)、复苏组(B组)、补阳还五汤饮片组(C组)、补阳还五汤颗粒剂组(D组)。建立窒息型大鼠心肺复苏模型,各组于自组循环恢复后24 h取样;采用双抗体夹心免疫吸收分析法(ELISA)测定血清和脑组织匀浆液NSE的水平并进行比较。结果:与假手术组比较,复苏组和各药物组大鼠血清NSE显著升高;与复苏组比较,各药物组大鼠血清NSE显著下降,但各药物组间差异无统计学意义;与假手术组比较,复苏组和各药物组大鼠脑组织NSE显著升高,与复苏组比较,各药物组脑组织匀浆NSE显著下降,但各药物组间差异无统计学意义。结论:NSE可作为心肺复苏后判断脑损伤的标志。大鼠心肺复苏后血清和脑组织匀浆NSE浓度升高。补阳还五汤饮片和颗粒剂能在不同程度上改善脑损伤指标,具有一定的脑保护作用,两者之间差异无统计学意义。 Objective: To observe NSE (Neuron Specific Enolase) serum and brain tissue homogenate levels by using a model of rat cardiac arrest after suffocation. Intervened by different formulations of Buyang Huanwu Decoction ( BYH- WD).,to research the effect of NSE brain tissue homogenate and serum levels in rats post cardiopulmonary resuscitation (CPR). Methods: 32 adult male Sprague - Dauley (SD) rats were divided into control group of Artificial operation (group) , recovery group ( group B) , decoction pieces of BYHWD group ( C ) , granule of BYHWD group ( D ) by ran- dom. Establishing the model of rat cardiac arrest after suffocation, all groups were sampled 24 b after autonomous cycle recovered, evaluating and comparing NSE brain homogenate and serum levels by double antibody sandwich immune ab- sorption process (ELISA). Results: The serum level of NSE was increased obviously in the two drug groups as well as in the recovery group compared with control group, and reduced significantly in the two drug groups compared with recovery group, but there was no significant difference between the drug groups. Conclusion: The NSE level may play as a sign of judgment of brain damage after cardiopulmonary resuscitation (CPR). The NSE serum and brain homogenate levels in rats" are increased after cardiopulmonary resuscitation. Both decoction pieces and granule of Buyang Huanwu Decoction (BYHWD) can improve brain damage index in different extents, and have certain protective effect, the difference be- tween the two has no statistical significance.
