高效液相色谱法同时测定玫瑰花口服液中没食子酸、芦丁、槲皮素和山奈素 被引量:3

Simultaneous Determination of Four Effective Components in Rose Oral Liquid by High Performance Liquid Chromatography

摘要 建立了同时测定玫瑰花口服液中没食子酸、芦丁、槲皮素和山奈素的高效液相色谱(HPLC)测定方法。采用Agilent HC-C18色谱柱(250×4.6mm,5μm);流动相为0.5%磷酸-甲醇,梯度洗脱;流速0.8mL·min-1;柱温30℃;检测波长260nm。结果显示:没食子酸在40.7~244.20μg·mL-1、芦丁在25~150μg·mL-1、槲皮素在13.3~80μg·mL-1、山奈素在2.69~16.2μg·mL-1线性关系良好,相关系数分别为0.9997、0.9993、0.9998、0.9994。4组分平均回收率分别为99.2%、101.2%、98.8%、99.2%,相对标准偏差(RSD)分别为0.60%、1.02%、0.62%、0.57%(n=3)。该方法准确、简便,适合上述4种有效成分的同时测定。 An analytical method for the simultaneous determination of four effective components(Gallic acid,Rutin,Quercetin,Kaempferol) in Rose Oral Liquid was established using high performance liquid chromatography(HPLC). The separation was performed on an Agilent C18 column(250 ×4.6 mm, 5 μm) at 30 C and 0.5 % phosphoric acid methanol as mobile phases with gradient elution at a flow rate of 0.8 mL/min,and the detection wavelength was 260 nm. Good linearity was obtained between the peak area and injected amount when the amount was 40.7-244.2 μg·mL-1 for Gallic acid,25.0-150.0 μg·mL-1 for Rutin,13.3 80 μg· mL 1 for Quercetin and 2.69-16.2 μg·mL-1 for Kaempferol. The correlation coefficient of each component was 0. 9997,0. 9993,0. 9998 and 0. 9994, respectively. Average recoveries of the four components were 99.2% ,101.2%,98.8% ,99.2%. RSD of each group were 0.60% ,1.02%, 0.62%,0.57% (n=3). Four effective components in Rose Oral Liquid were simultaneously determined, and the method was convenient and accurate.
