缺氧诱导人肺癌细胞株表达血管内皮生长因子 被引量:1

摘要 目的 检测缺氧对人肺癌细胞株表达 VEGF的影响及其机制。方法  4株人肺癌细胞在低氧环境下处理 18h后 ,运用半定量 RT- CPR及免疫组化技术分别检测 VEGFm RNA 及蛋白的表达。结果 低氧处理后人肺癌细胞VEGF m RNA明显高于未处理之肺癌细胞并在免疫组化上得到了进一步证实。 Objective To investigate the influence hypoxia on the expression of vascular endothelial growth factor (VEGF) expression and its mechanism.Methods The expression of VEGF mRNA and protein in four hypoxia-treated human lung cancer cell lines (A549,A1,A7,A8) were tested determined by using semiquantitative reverse transcription PCR (RT-PCR) and immunohistochemcal staining. Results Semiquantitative RT-PCR demonstrated that the amount of VEGF mRNA in cells treated by hypoxia increased markedly when compared with that of control lines and the above results was confirmed by immunohistochemical staining. Besides, 3 VEGF isoforms(VEGF 121,165 and 189) could also be detected by semiquantitative RT-PCR.Conclusion These data that VEGF expression may be up-regulated by hypoxia.
