摘要
从造纸厂碱性土样中分离到 2 4株能够产生纤维素酶的真菌 ,经过摇瓶复筛 ,选出一株耐碱性纤维素酶产生菌S60 7。经过鉴定 ,该菌株为黑色葡萄状穗霉 (Stachy botrysatra)。通过对其粗酶液的性质进行测定 ,该菌所产纤维素酶粗酶液中的FPA、CMCase和β-葡萄糖苷酶的最适pH分别为 6.0 ,6.0和 6.5 ,其中内切酶在pH 9.0时仍能保持最高酶活力的 5 0 %以上 。
Twenty four strains of fungi producing cellulases were isolated from alkaline soil on the site of paper mill. One of them being identified as Stachybotrys atra was chosen to be further studied because its cellulases had good endurance in alkaline condition. The optimum pHs of the FPase, CMCase and β-glucosidase activities in the crude enzyme were 6.0, 6.0 and 6.5 respectively. More than 50% of endoglucanase (CMCase) activity could be obtained at pH 9.0. The results showed that the cellulase from the strain has high pH and temperature stabilities.
