聚合酶链式反应及单链构象多态性技术作为一种区分检测基因组之间微小差异的有效方法 ,具有快速、简便、灵敏和适用于大样品量筛选的特点 .本文详细介绍了该技术的条件优化及改进策略 .该技术在植物病毒学上主要应用于 :(1)分子变异 ;(2 )混合侵染的检测 ;(3)
Polymerase chain reaction single strand conformation polymorphism (PCR SSCP) technique is a method feasible to detect minor sequence changes in PCR amplified DNA. Because of its rapidness, simplicity, sensitivity and applicability to large scale screening, it has been applied to plant virology including molecular variability, detecting mixed infections and molecular epidemiology. Its conditions optimization and improvement are reviewed in detail.
