RAPD Analysis of Several Rabbit Lines

摘要 应用随机扩增多态 DNA技术 ( RAPD)对新西兰白兔、加利福尼亚兔、布列塔尼亚配套系( ELCO)进行了遗传分析。从 2 0个随机引物中筛选出 8个 ,对其基因组 DNA进行 PCR扩增 ,对扩增产物进行琼脂糖凝胶电泳 ,共检测到 71条扩增片段 ,其中多态片段 45条 ,占 63.4%,反映了家兔品种的遗传变异。多态性统计表明 :品种内的遗传相似度大于品种间的遗传相似度。聚类分析表明 :ELCO的 D系与新西兰白兔、加利福尼亚兔的亲缘关系较近 ,聚为一个类群 ;ELCO的 A系与B系亲缘关系较近 。 Randomly Amplified Polymorphic DNA (RAPD) technique was used to analyse genetic relationships among New Zealand White rabbits, California rabbits and 4 lines of ELCO (A, B, C, D). 8 primers screened from 20 random primers were used to amplify the genomic DNA of rabbits. Seventy-one amplified fragments were detected by electrophoring with agarose. Among them, 45 fragments were polymorphic, accounting for 63.4% of the whole. It was showed that the RAPD polymorphisms among rabbit lines were abundant. The results of the statistics and analysis of RAPD bands of rabbits showed that the genetic similarity within the lines was higher than which among the lines. The cluster analysis showed that the relationship among D line of ELCO, New Zealand White rabbits and California rabbits, which classified one group, was nearer than the others. A line and B line of ELCO were classified one group. This study indicated that RAPD technique could be used to analyse genetic variations among within rabbit lines. It was an effective marker to estimate the group genetic relationships among rabbits, and could give theoretical and research basis for line selection and crossbreeding of rabbits.
