MicroRNA-29b在食管鳞状细胞癌中的表达及临床意义 被引量:8

摘要 目的探讨MicroRNA-29b(miR-29b)在食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)组织中的表达情况,并分析miR-29b与食管鳞癌临床病理特征及预后之间的关系。方法采用实时荧光定量聚合酶链反应法(real-time quantitative PCR,qPCR)检测84例ESCC组织和癌旁正常组织中miR-29b的表达差异,并分析miR-29b表达情况与食管鳞癌患者的临床病理特征、预后的相关性。结果 miR-29b不同程度低表达48例(57.2%),正常表达28例(33.3%),高表达8例(9.5%)。不吸烟患者癌组织中miR-29b相对表达水平低于吸烟者(P<0.05);miR-29b相对低表达患者的肿瘤浸润程度低,TNM分期早(P<0.05);miR-29b相对低表达患者的无瘤生存期较非低表达患者长(P<0.05);miR-29b表达情况和肿瘤家族史是影响食管鳞癌患者生存的独立因素(P<0.05)。结论 miR-29b相对正常组织低表达能减缓食管癌进展和改善预后,miR-29b低表达可能起着抑癌因子的作用,检测miR-29b表达情况可能对食管癌进展和预后具有一定临床意义。 Objective To detect the expression of miR-29b in esophageal squamous cell carcinoma (ESCC) and to analyze its correlation with clinicopathological features and prognosis of ESCC. Methods The expression of miR-29b in 84 ESCC samples and matched normal tissue samples was detected by real-time PCR. The relationship between the expression and elinicopatbological features and prognosis of ESCC was analyzed. Results Forty-eight cases (57.2%) of carcinoma samples showed low expression of miR-29b, which was correlated with tumor infiltrating degree and pathological staging in ESCC (both P 〈 0.05 ), while 28 cases (33.3%) were negative and 8 (9.5%) showed high expression. The disease-free survival in patients with low expression of miR-29b was longer than that in those with non-low repression( P 〈 0.05 ) , The expression of miR-29b and tumor family history was one of the factors affecting the survival of patients with esophageal squamous cell carcinoma ( P 〈 0.05 ). Conclusions The low expression of miR- 29b may participate the progression and prognosis of esophageal squamous cell carcinoma as a tumor suppressor factor. Detection of miR-29b expression may have certain clinical significance on the progression and prognosis of esophageal squamous cell carcinoma.
