摘要
由屠宰母猪卵巢分离得到505枚卵母细胞,经成熟培养,其中468枚卵丘细胞扩张,成熟率为92.7%。采用冷冻的附睾精子授精,受精率为41.9%(57/136),其中单精受精率36.8%(50/136),多精受精率5.1%(7/136)。授精卵体外培养24和48小时后的卵裂率分别为5.3%(15/258)和12.9%(30/232)。把35枚早卵裂期胚胎和250枚未见卵裂的1-细胞期受精卵移入4头受体母猪的输卵管内,其中2头妊娠,分别于1990年4月10日和29日正常分娩,共生产仔猪21头。
This study was undertaken to establish a system that in vitro maturation pig follicular oocytes were fertilized in vitro by frozen boar epididymal spermatozoa. Five hundred and five pig follicular oocytes with compact cumulus cells and qualified vitelli were obtained from the ovaries collected in a local slaughterhouse. Ninety two point seven percent (468/505) of the oocvtes matured 36 hrs after cultured ih medium TCM-199 with 5 mg/ml BSA and 10 IU/ml PMSG. Forty two percent (57/136) of oocytes matured in vitro were fertilized 8 hrs after inseminated in BO solution containing 10 mg/ml BSA and 2 mM-caffeine with the frozen epididymal spermatozoa capacitated in medium TCM-199 supplemented with 0.9 mg/ml calcium lactate, 0.1 mg/ml sodium pyruvate, 0.55 mg/ml D-glucose and 12% FCS. The percentage of mono-spermic oocytes and polyspermic oocytes were 36.8% (50/136) and 5.1% (7/136) respectively. Five percent (15/285) and 12.9% (30/232) of inseminated oocytes reached 2- to 4-cell stage 24 and 48 hrs respectively after cultured in a droplet of TCM-199 with 10% FCS under paraf-fine oil at 39℃, 5% CO2 in air with maxium humidity. Thirty five embryos (2- to 4-cell stage) and 250 inseminated oocytes were surgically transferred to the oviducts of 4 recipients. Two of them became pregnant. One farrowed 8 piglets and another did 13 piglets. In conclusion, the results showed that offsprings could be produced by the system of in vitro fertilization set up in this study.
