摘要
研究了不同激素配比、性周期阶段对白牦牛卵母细胞体外成熟的影响,同时研究了不同培养基对早期胚胎体外发育的影响。结果表明:M199+0.2 mmol/L丙酮酸钠+10%NBS+5.0 mg/L LH+0.5 mg/L FSH+1μg/ml 17β-E2是白牦牛卵母细胞体外成熟较为理想的培养系统(成熟率79.4%、卵裂率42.5%)。采集白牦牛卵母细胞时卵巢所处性周期阶段(卵泡期、黄体期)影响卵母细胞的体外成熟(成熟率分别为77.6%、69.5%,卵裂率分别为45.7%、35.6%)。输卵管上皮细胞和颗粒细胞是理想的共培养细胞,能有效克服白牦牛早期胚胎发育阻滞(桑囊胚发育率分别为10.6%、7.7%)。
Effect of different hormone composition and estrous cycle on maturation in vitro for oocytes of white yak was studied. At the same time, effect of different culture medium on development in vitro for earlier embryo of white yak was researched. The results showed that M199+0. 2 mmol/L Na-pyruvate+10%NBS +5.0 mg/L LH+0. 5 mg/L FSH+1 μg/ml 17β-E2 was ideal cul ture system for in vitro maturation of white yak oocytes (the maturation rate was 79.4%, and the cleavage rate was 42. 5 %). Oocytes collected from ovaries in different estrous cycle (follicular phase, luteal phase) had significant difference in vitro maturation of white yak oocytes ( the maturation rates were 77.6% and 69.5%; and the cleavage rates were 45.7% and 35.6 %, respectively). Bovine oviduct epidermic cell and granulosa cell were all ideal co-culture cells, which could effectively surmount development block of earlier embryo of white yak (the development rates were 10.6 % and 7.7%, respectively).
