PrP^C蛋白对小鼠学习与记忆能力的影响 被引量:1

The role of PrP^C protein in the mouse learning and memory ability

摘要 利用本实验室构建的C57BL/6背景下的PRNP基因敲除小鼠模型与C57BL/6野生型小鼠,通过避暗试验以及Morris水迷宫试验观察小鼠的行为。以此来研究PrPC蛋白的缺失对于小鼠学习与记忆能力影响,进而反映PrPC蛋白对于神经系统功能的影响。结果显示,PRNP敲除小鼠在避暗试验中对于电刺激的记忆比野生型小鼠强,且在第3天差异显著(P<0.05)。在Morris水迷宫定位航行试验中,敲除鼠的逃避潜伏期明显长于野生小鼠,第3、5、6天差异显著(P<0.05)。空间探索试验中,PRNP敲除鼠与野生小鼠相比较,首次穿越虚拟平台的时间以及穿越平台的次数差异并不显著。研究表明,PrPC蛋白缺失对于小鼠主动回避伤害的记忆能力有所提高,但是对于空间立体学习和记忆能力损害很大。 In order to study the absence of PrPC protein for the mouse learning and memory ability,thus reflect the impact of the PrPC protein for nervous system function.We observed the behavior of PrP0/0 mice and C57BL/6 wild-type mice in the passive avoidance test and Morris water maze test.In passive avoidance experiments,compared with wild type mice,the PrP0/0 mice were better to avoid the electrical stimulation,especially on the 3th(P0.05).The escape latency of PrP0/0 mice was significantly longer than wild-type mice in the Morris water maze experiment,especially on the 5th and 6th(P0.05).In PrP0/0 mice,the time of first crossings platform location and the number of crossings platform location were no significant difference when compared with C57BL/ 6 mice in the locus of probe trial test.That of PrPC protein deficient mice active avoidance of injury and memory ability improved,but is damage to the spatial learning and memory ability.
