摘要
研究比较了CTAB法和SDS法在枣和酸枣基因组DNA提取中的效果 ,并分析了取样时间、部位、样品状况、抗氧化剂和不同纯化处理等对所提DNA质和量及其RAPD扩增效果的影响。结果表明 :用改良后的CTAB法优于SDS法 ,可有效地去除多糖 ,所提DNA的质和量均能满足PCR扩增要求。取样时间与部位对所提基因组DNA的质量无影响 ,但与产量有关 ,以旺盛生长期的幼叶或嫩梢尖为佳。样品采后液氮处理 - 70℃低温保存 ,与新鲜材料所提DNA差异不大。抗氧化剂可有效地阻止多酚类物质氧化变褐 ,1%的 β 巯基乙醇即可满足要求。
WT5BZ]The effects for extracting genomic DNA by the methods of CTAB and SDS were studied on Chinese jujube and acid jujube, and the effects of different sampling period, position, storage treatment, and oxidation inhibitor etc., on the quality and quantity of DNA extracted and RAPD results were compared.The results showed that the improved CTAB method was better than that of the SDS method; it could remove polysaccharide effectively.The sampling period and position had no effect on the quality of DNA extracted, but they had relation to the quantity of DNA extracted. The young leaf or shoot was good for DNA isolation. The sample treated with liquid nitrogen and stored with -70℃ refrigerator had no difference with fresh sample. The browning of DNA could be prevented by 1% β mercaptoethanol. RNA and a little protein had no effect on RAPD results.
