摘要
目的 研究白血病细胞耐药性与Bcl-2和Bax -alpha阳性率及表达水平关系。方法 选择难治或复发的化疗耐药白血病AML -2患者和初治化疗敏感白血病AML -2患者各 10例 ,分离白血病细胞 ,应用流式细胞术比较分析化疗耐药白血病细胞与化疗敏感白血病细胞内Bcl-2和Bax -alpha阳性率和表达水平的关系。结果 耐药组和敏感组中Bcl -2细胞的阳性率和相对荧光强度均无统计学差异 ;而Bax -alpha阳性率在耐药组和敏感组分别为 (11 2 7±13 2 5 ) %和 (6 5 39± 30 0 1) % ,两组间差异有显著性 (P =0 0 0 1) ,而相对荧光强度分别为 4 5 9± 4 6 0和 7 89± 3 5 2 ,无统计学差异 (P =0 0 78) ;两组间Bcl-2∶Bax -alpha比率分别为 12 14± 10 70和 2 79± 1 43 ,差异具有显著性 (P =0 0 16 )。结论 Bax -alpha的表达水平和Bcl-2和Bax
Objective To study the correlation between leukemia chemoresisance and the level of Bcl-2 or Bax-alpha expression.Methods Patients were divided into two groups: 10 cases of chemoresistant leukemia (refract or relapsed leukemia, AML-2) and 10 cases of chemosensitive leukemia (primary treated sensitive leukemia, AML-2). Multiparameter flow cytometry was used to measure the percentages and intensity of Bcl-2 and Bax-alpha expression by leukemia cells.Results The level and intensity of Bcl-2 expression did not have significant differences between both groups. The percentages of Bax-alpha positive leukemia cells in chemoresistance group and chemosensitive groups were with significant difference ( P <0 01). And the levels of Bax-alpha expression were 4.59±4.60 and 7.89±3.52 respectively ( P =0 078). The Bcl-2/Bax-alpha ratio were significantly and chemosensitive group ( P =0 016), they were (12.14±10.70) vs (2.79±1.43). Conclusion Bax-alpha expression and Bcl-2/Bax-alpha ratio may be important in defermining on chemoresponse of leukemia cells.
