肺癌组织中Endostatin mRNA转录表达与血清Endostatin水平关系的相关性研究 被引量：4

Correlation between Endostatin Serum Levels and Endostatin mRNA Transcription Levels in Lung Cancer Tissues

摘要 目的:探讨非小细胞肺癌患者肺癌组织中EndostatinmRNA转录表达水平与血清中内皮抑素(Endo-statin)水平的关系以及两者水平与肺癌临床病理生理特征的关系。方法:用ELISA法检测46例非小细胞肺癌患者血清中Endostatin的水平;用RT-PCR法检测其肺癌组织中EndostatinmRNA的转录表达水平。结果:1)肺癌患者血清中Endostatin水平为20.85±4.56ng/ml;肺癌组织中EndostatinmRNA的转录表达产物的OD比值为0.872±0.071。2)肺癌患者血清中Endostatin水平及肺癌组织中EndostatinmRNA的转录表达水平均与肺癌原发肿瘤大小、有无远处转移、细胞分化程度和P-TNM分期等有密切关系(P<0.05),而与肺癌原发部位、淋巴结转移状态、组织学类型,患者性别、年龄和吸烟与否等均无明显关系(P>0.05)。3)肺癌组织中CollagenⅩⅧ/EndostatinmRNA的转录表达与血清中Endostatin水平呈非常显著正相关(r=0.686,P<0.01)。结论:肺癌患者肺癌组织中EndostatinmRNA的转录表达与血清Endostatin水平有非常显著正相关关系,且均与肺癌的发生、发展、远处转移和细胞分化不良有密切关系,非小细胞肺癌患者血清中Endostatin水平和肺癌组织中EndostatinmRNA的转录表达水平可能是预测肺癌恶性行为的有用指标。 Objective： To study the correlation between the serum level of endostatin and the expression of Collagen ⅩⅧ/Endostatin mRNA in lung cancer tissues and to explore the relationship between these levels and the clinical and pathophysiological characteristics in non-small cell lung cancer （NSCLC） patients. Methods： The serum level of endostatin was detected in 46 patients with NSCLC using ELISA and the expression level of Collagen ⅩⅧ/Endostatin mRNA was detected in the lung cancer tissues by RT-PCR. Results： a） The mean serum concentration of endostatin in the lung cancer patients was 20.85±56ng/ml and the expression level of Collagen ⅩⅧ/Endostatin mRNA in the lung cancer tissues was 0.872±071. b） The serum concentration of endostatin and the transcription expression of Collagen ⅩⅧ/Endostatin mRNA were closely related to the P-TNM stages, presence of distant metastasis, grade of cell differentiation and the size of the primary tumor （P〈0.05）, but not to the histological classification, type of tumor, lymph node status, age, sex of the patients or cigarette use （P〉 0.05）. c） The serum level of endostatin in lung cancer patients had a highly positive correlation to the expression of Collagen ⅩⅧ/Endostatin mRNA in lung cancer tissues （r=0.686, P〈0.01）. Conclusion：The serum level of endostatin in lung cancer patients has a highly positive correlation to the expression of Collagen ⅩⅧ/Endostatin mRNA in lung cancer tissues.The serum level of endostatin and the expression of Collagen ⅩⅧ/Endostatin mRNA in patients wiht NSCLC might be helpful for evaluating the biological behavior of lung cancer.
