摘要
利用酪蛋白培养基从猪粪便样品中分离产蛋白酶的芽胞杆菌,采用福林酚法测定其酶活力,对筛选到的高酶活菌株进行形态学、生理生化和分子鉴定。结果表明,从86株分离菌中筛选了一株酶活较大的菌株B.A464,革兰氏阳性菌,呈粗杆状,能形成卵圆形芽孢,16S rDNA序列长度是1 463 bp,与蜡样芽孢杆菌的同源性高达99%,鉴定为蜡样芽孢杆菌,所产蛋白酶活力为2.19×103U·mL-1。
Bacillus producing proteinase were isolated from pig feces by medium containing casein as sub- strate, the proteinase activities of above bacteria were detected by Forint phenol method, and the high proteinase- producing Bacillus were screened. They were identified by the colony morphology, microscopic morphology in com- bination with the physiological and biochemical feature and 16S rDNA sequence. The results showed that the strain B.A464 with the highest proteinase activity of 2.19×10^3U·mL^-1, was obtained, and it was gram strain positive, short rod, forming oval spores, and had the ability of fermenting many sugars. Based on the 16S rDNA sequence homology, comparative analysis showed that it belonged to the genus bacillus and most closery related to bacillus cereus strain 08001 with 99% sequence simmarity. So the strain B.A464 was identified to be bacillus cereus.
