摘要
目的观察水凝胶角膜给药载体对大鼠角膜炎的治疗效果。方法建立金黄色葡萄球菌角膜炎动物模型,SD大鼠随机分为5组:分子印迹聚甲基丙烯酸-β-羟乙酯与甲基丙烯酸的共聚物P(HEMA-CO-MAA)水凝胶角膜给药载体组、聚甲基丙烯酸-β-羟乙酯与甲基丙烯酸的共聚物P(HEMA-CO-MAA)水凝胶角膜给药载体组、聚甲基丙烯酸-β-羟乙酯(PHEMA)水凝胶角膜给药载体组、加替沙星眼药水组、自愈组,每组各3只,分组进行饲养。比较各组角膜结膜情况并进行角膜细菌划线培养和病理组织学观察。结果角膜炎大鼠干预后48 h进行检测,自愈组划线细菌培养与干预前无明显差别,PHEMA组划线菌落培养少于干预前,而加替沙星眼药水组、P(HEMA-CO-MAA)组及分子印迹P(HEMA-CO-MAA)组划线菌落培养明显少于干预前。角膜冰冻切片行HE染色,各组均可见水肿,相对于眼药水组,P(HEMA-CO-MAA)及分子印迹P(HEMA-CO-MAA)水凝胶角膜给药载体组角膜基质水肿程度较弱,炎症浸润细胞较少。结论 P(HEMA-CO-MAA)和分子印迹P(HEMA-CO-MAA)水凝胶角膜给药载体比PHEMA水凝胶角膜给药载体杀菌效果强,更具有未来长效缓释的研究价值。
Objective To observe the effect of hydrogel drug delivery on keratitis in rats.Methods The animal model of Staphylococcus aureus-induced keratitis was established in rats.Animal models were randomly divided into five groups including the imprinted P(HEMA-CO-MAA) hydrogel corneal for drug carrier group,the copolymerized P(HEMA-CO-MAA) hydrogel corneal for drug carrier group,the PHEMA hydrogel corneal for drug carrier group,the gatifloxacin eye drops group and the self-healing group.Each group contained 3 rats and fed separately.The situation of cornea conjunctiva,corneal bacteria culture and pathological histology were observed.Results The bacterial culture in the self-healing group was no statistical difference before and after intervention at 48 h.The bacterial colony in the PHEMA group was obviously reduced after intervention at 48 h.The bacterial colonies in the gatifloxacin eye drops group,the copolymerized P(HEMA-CO-MAA) group and the imprinted P(HEMA-CO-MAA) group were significantly decreased after intervention at 48 h.Compared with the gatifloxacin eye drops group,the corneal stromal edema and inflammatory cell infiltration were diminished in the copolymerized P(HEMA-CO-MAA) and imprinted P(HEMA-CO-MAA) groups.Conclusion Copolymerized P(HEMA-CO-MAA) and imprinted P(HEMA-CO-MAA) hydrogel corneal for drug delivery systems are superior to PHEMA hydrogel corneal for drug carrier in bactericidal effect,which is related to the sustained release.
