摘要
目的 :观察三氧化二砷 (AS2 O3)对肺癌细胞株NCI H6 9的增殖抑制作用并初步探讨其机制。方法 :肺癌细胞株NCI H6 9用不同浓度AS2 O3处理。MTT法观察不同浓度AS2 O3作用不同时间后对NCI H6 9生长的影响 ;增殖细胞核抗原 (PCNA)单抗LSAB免疫组化染色 流式细胞仪检测PCNA阳性细胞百分比。结果 :AS2 O3明显抑制肺癌细胞株NCI H6 9细胞增殖 ,其作用随AS2 O3浓度增加和作用时间延长而增强 ;随AS2 O3浓度增加 ,PCNA阳性细胞百分比逐渐下降。结论 :AS2 O3可明显抑制肺癌细胞株NCI H6 9的生长 ,调控DNA合成及细胞增殖的PCNA蛋白合成降低可能是AS2 O3的作用机制之一。
Purpose:To observe the effect of proliferation inhibition of Arsenic Trioxide(AS 2O 3) on lung cancer cell line NCI H69.Methods:Lung cancer cell line NCI H69 was treated with different concentrations of AS 2O 3. MTT assay was used to observe the proliferation status. Flow cytometry was used to measure the percentage of proliferating cell nuclear antigen (PCNA) positive cell. Results:AS 2O 3 could significantly inhibit the growth of NCI H69 cell, this effect was dose and time dependent. The percentage of PCNA positive cell was decreased gradually when concentration of AS 2O 3 was increased. Conclusions:Arsenic Trioxide could inhibit the proliferation activity of lung cancer cell line SW620. Reduction of PCNA which was related to the regulation of DNA synthesis and cell proliferation may be the mechanism of this proliferation inhibition.
