三氧化二砷通过自噬作用抑制A549肺癌细胞的生长 被引量:3

摘要 目的探讨三氧化二砷对肺癌细胞生长的影响,揭示三氧化二砷抗癌作用的机制。方法培养人肺癌细胞A549,给予不同药物浓度三氧化二砷药物处理;利用CCK8检测三氧化二砷对A549细胞活力的影响;以自噬试剂盒观测细胞的自噬情况。结果 10、25、50和100μmol/L三氧化二砷明显抑制A549的细胞活力(P<0.05)。三氧化二砷处理后镜下见阳性自噬细胞增多,自噬率增加。结论 AS2O3可抑制A549肺癌细胞生长,这种效应可能通过诱导细胞自噬来发挥作用。 Objective To investigate the role of arsenic trioxide on growth inhibition and its mechanisms. Methods The lung cancer cell A549 were cultured and treated with different concentration of arsenic trioxide. CCK-8 assay was used for evaluing the cell viability of A549 cells with or without treatment. Autophagy detection kit was used for investgating the status of autophagy. Results 10, 25, 50, 100μmol/L arsenic trioxide significantly inhibit the proliferation ( P 〈 0. 05 ) in A549 cells. Increased autophagy cells is photographed under microscope in arsenic trioxide treated cells. Rate of autophagy cell is higher in arsenic trioxide treated ceils than that in untreated cells. Conclusion Arsenic trioxide inhibit the growth of lung cancer cells partly through increasing the cell autophagy.
