成骨生长肽调节鼠胚颅盖骨成骨细胞样细胞酶活性的观察 被引量:4

摘要 目的 观察成骨生长肽 ( Osteogenic growth peptide,简称 OGP)对 SD大鼠鼠胚颅盖骨成骨细胞样细胞酶活性的影响。方法 无菌条件下培养鼠胚颅盖骨成骨细胞样细胞 ,分为不加 OGP的对照组和加 OGP的用药组 ,用药组在 10 -12 M~ 10 -8M OGP浓度范围内 ,再分为三组 (分别为 OGP浓度10 -12 M、10 -10 M、10 -8M) ,收集以上四组细胞及细胞培养液 ,测定它们的碱性磷酸酶及酸性磷酸酶活性。结果 用药组的碱性磷酸酶活性均显著地高于对照组 ( P<0 .0 1~ 0 .0 5,用药组 OGP10 -8M浓度、培养7天的细胞培养液中碱性磷酸酶活性为 0 .82± 0 .16U/ L,而对照组同样条件活性为 0 .2 3± 0 .10 U/ L) ,且随着 OGP浓度的升高碱性磷酸酶活性也升高 ,而酸性磷酸酶用药组与对照组无显著差异。 Objective\ To investigate the regulatory role of osteogenic growth peptide (OGP)on enzyme activity of osteoblast like cells in rats.Methods\ The isolated osteoblastic cells from calvaria of newborn ras were cultured in vitro. Cutured osteoblastic like cells were divided into two groups:one group was added with OGP,the other was added with DMEM as the control group.The experimental group was further divided into three subgroups(10 -8 M,10 -10 M and 10 -12 M OGP).The activities of alkaline phosphatase and acid phosphatase in these 4 groups were determined.Results The activities of alkaline phosphatase in cells and extracellular matrix of the experimental group were significantly higher than that of the control group( P <0.01,0.82±0.16 U/L for OGP 10 -8 M group on 7 th culture day in extracellular matrix and 0.23±0.10 U/L for the control group).No change in activities of acid phosphatase in osteoblast like cells and extracellular matrix between the control group and the experimented group was observed.The alkaline phosphatase activities of cells and extracellular matrix in the exprimental group were gradually upregulated with increasing concentrations of OGP from 10 -12 M to 10 -8 M in range. Conclusion OGP can enhance alkaline phosphatase activity in osteoblast like cells,thereby promoting osteogenic effect.
