ghrelin在致渴条件下对大鼠饮水的影响

摘要 目的探讨ghrelin在致渴条件下对水食摄入的调节。方法给予大鼠以下5种致渴刺激:①禁水36h;②用30g/L氯化钠代替饮用水;③外周注射高渗盐水;④侧脑室(ICV)注射血管紧缩素Ⅱ(AngⅡ);⑤外周注射高渗盐水+中枢注射AngⅡ。之后大鼠经ICV注射ghrelin0.4μg,观察大鼠摄食摄水量的改变。结果禁水36h后ghrelin可显著增加大鼠摄食量(t=4.900,P<0.05);饮用36h高渗盐水后ICV注射ghrelin大鼠摄食量显著增加(t=6.018,P<0.01);外周注射高渗盐水后ICV注射ghrelin大鼠摄食量显著增加(t=8.133,P<0.01),但摄水量显著减少(t=4.256~6.382,P<0.01);ICV注射AngⅡ大鼠摄食量无明显变化(P>0.05),但摄水量显著增加,ghrelin可显著降低由AngⅡ所致的饮水量增加(F=17.66~87.32,P<0.01);外周注射高渗盐水+中枢注射AngⅡ后,ghrelin可显著增加大鼠摄食量(t=6.348,P<0.05)。结论 ghrelin的作用并不仅限于摄食调节,也参与摄水调控。 Objective To study the effects of ghrelin on water intake under dipsogenic condition. Methods The rats were exposed to one of the following five dipsogenic stimuli: ①water deprivation 36 h; ② drinking water replaced by 30 g/L NaCl; ③peripheral injection of hypertonic saline; ④ ICV injection of angiotensin Ⅱ fang Ⅱ) ; ⑤ peripheral injection of hyperton ic saline+central Ang Ⅱ. The rats were then given ICV injection of ghrelin (0.4 t^g), the changes of food and water intake were observed. Results After 36-hour water deprivation, ghrelin notably increased food intake (t =4.900,P〈0.05) ; After drinking hypertonic saline for 36 hours followed by ICV injection of ghrelin, food intake dramatically increased (t= 6.018,P〈0.01) ; After peripheral injection of hypertonic saline followed by ICV injection of ghrelin obviously increased food intake (t = 8.133, P〈0.01), but water intake markedly decreased (t=4.256-6.382,P〈0.01). ICV injection of AngⅡ did not change the food intake, but in- crease the waler intake, which could be lowered with ghrelin injection (F=17.66-87.32,P〈0.01). After peripheral injection of hypertonic saline plus central injection of AngⅡ , ghrelin could significantly increase the food intake (t =6.348,P〈0.05). Con- clusion Ghrelin involves not only in regulation of food intake, but also in water intake.
