白藜芦醇对人肝癌细胞转移的影响及其分子机制 被引量:8

摘要 目的观察白藜芦醇的抗肿瘤活性,并探讨其抑制抗血管发生及肿瘤转移的分子机制。方法体外培养HepG2细胞,用12.5、25、50、100μmol/L的白藜芦醇刺激。小室侵袭实验检测HepG2细胞的迁移与侵袭;Western blot法检测基质金属蛋白酶(MMP)-2、MMP-9、血红素氧合酶1(HO-1)的表达以及NF-κB p65亚基核转位。结果 50μmol/L白藜芦醇孵育24 h后,其迁移与侵袭率分别降低了37%和33%;Western blot结果显示白藜芦醇能抑制p65核转位;此外,抑制并沉默HO-1表达后,能显著降低MMP的表达。结论白藜芦醇对MMP的表达影响很大程度上与抑制HO-1-NF-κB信号通路活性有关。 Objective To observe the anti-carcinogenic activities of resveratrol,and then investigate the molecular mechanism involved in angiogenesis and tumor metastasis.Methods Hepatoma carcinoma cell line HepG2 was cultured in vitro,and stimulated by 12.5,25,50,100 μmol/L of resveratrol.The migratory and invasive abilities were assessed by chamber invasion assay.Expression of matrix metalloproteinase-2,9 and heme oxygenase-1(HO-1),translocation of NF-κB p65 subunit were detected by Western blot.Results After treatment of HepG2 cells with resveratrol(50 mmol/L) for 24 h,the migratory and invasive abilities(37% and 33% inhibition,respectively) were significantly reduced.Western blot showed resveratrol could inhibit p65 translocation.Furthermore,HO-1 inhibition and silencing significantly suppressed MMPs expression.Conclusion The inhibitory effects of resveratrol on MMP expression of hepatoma carcinoma cells are partly associated with the HO-1-mediated NF-κB pathway.
