摘要
目的观察六味地黄汤对5/6肾切除大鼠肾间质纤维化的拮抗作用和对肝细胞生长因子(hepatocyte growth factor,HGF)及其特异性受体c-Met表达的影响。方法 SD大鼠分为假手术组、模型组和六味地黄汤组,采用5/6肾切除法复制慢性肾间质纤维化模型。造模1周后,灌胃干预8周。观察残肾病理改变,采用免疫组化检测肾组织中HGF和c-Met蛋白的表达。结果模型组和六味地黄汤组残肾均出现肾间质纤维化,模型组更为严重;与假手术组相比,模型组HGF表达升高(P<0.05),六味地黄汤组表达也升高(P<0.01),六味地黄汤组HGF表达强于模型组(P<0.05);与假手术组相比,模型组和六味地黄汤组c-Met表达均升高(P<0.01),六味地黄汤组表达强于模型组(P<0.01)。结论六味地黄汤可能通过促进5/6肾切除大鼠肾组织HGF及其受体c-Met的表达,减轻肾间质纤维化程度。
Objective To observe the protection of Liuwei Dihuang Decoction on renal interstitial fibrosis,and to investigate its effects on hepatocyte growth factor （ HGF） and its receptor （ c-Met） in 5 /6 nephrectomy rats. Methods SD rats were divided into sham operation group,model group and Liuwei Dihuang Decoction group,and adopted 5 /6 nephrectomy to copied chronic renal interstitial fibrosis. A week after operation,rats were administrated every day for 8 weeks. Pathological changes of the remnant kidney were observed by HE and Masson staining,Immunohistochemistry was used to observe the expression of HGF and c-Met protein. Results the model group and the Liuwei Dihuang Decoction group both appeared renal fibrosis,the model group was more obvious; Compared with the sham operation group,the expression of HGF manifested obvious promotion in the model group （ P〈0. 05） ,the expression in the Liuwei Dihuang Decoction group raised too （ P〈0. 01） ,compared with the model group,the expression in the Liuwei Dihuang Decoction group were more serious （ P〈0. 05） ; Compared with the sham operation group,the expression of c-Met manifested obvious promotion in the model group and the Liuwei Dihuang Decoction group （ P〈0. 01） ,compared with the model group,the expression in the Liuwei Dihuang Decoction group manifested more serious （ P〈0. 01） . Conclusion Liuwei Dihuang Decoction may promote the expression of HGF and c-Met in 5 /6 nephrectomy rats and inhibit the development of renal interstitial fibrosis.
