高效液相色谱法测定姜黄素清蛋白纳米粒中药物含量与包封率 被引量:2

摘要 目的建立测定姜黄素清蛋白纳米粒中主药含量及包封率的方法。方法采用高效液相色谱法,色谱柱为Hypersil C18柱(250 mm×4.6 mm,5μm);流动相:乙腈-4%冰醋酸(55∶45);流速:1.0 mL.min-1;柱温:30℃;检测波长:425 nm;进样量:20μL;采用低温超速离心法分离姜黄素清蛋白纳米粒和游离药物,测定包封率。结果姜黄素在0.102~16.32μg.mL-1浓度范围内线性关系良好(r=0.999 9),平均回收率为(99.96±1.01)%(n=9),以标示量计平均含量为(100.31±0.94)%,平均包封率为(71.13±1.30)%。结论该方法灵敏、简便、准确,适用于姜黄素清蛋白纳米粒药物含量及包封率的测定。 Objective To establish a method for determining the content and entrapment efficiency of eureumin albumin nanopartieles. Methods The separation was performed on a Hypersil C ls column (250 mm×4.6 mm,5 μm) with a mobile phase of 4% glacial acetic aeid-aeetonitrile (45 : 55 ). The process was carried out with a flow rate of 1.0 mL ·min^-1 , column temperature at 30 ℃ and wavelength at 425 nm. Refrigerated high-speed eentrifugation method was employed to isolate free eureumin for the entrapment efficiency determination. Results The calibration curve was linear in the range of 0. 102- 16.32 μg · mL^-1(r=0.999 9). The mean recovery (n=9) was (99.96±1.01) %. The mean content of labeled amount was (100.31±0.94) % ,and encapsulation efficiency was (71.13±1.30) %. Conclusion The method is sensitive,simple, accurate and applicable for determining entrapment efficiency of eureumin in the albumin nanopartieles.
