摘要
目的研究附子理中汤对腹泻型肠易激综合征(D-IBS)大鼠血清肿瘤坏死因子α(TNF-α)以及白介素10(IL-10)的影响,初步探讨附子理中汤治疗D-IBS的药理学基础。方法 50只大鼠随机分为正常对照组、模型组、得舒特组、附子理中汤大、小剂量组;除对照组外各组均采用番泻叶加束缚应激构建大鼠D-IBS模型,2周后予相应的药物治疗;采用酶联免疫吸附测定法(ELISA)测定各组大鼠血清TNF-α、IL-10水平。结果模型组大鼠血清TNF-α水平较正常对照组高(P<0.05),各药物干预组血清TNF-α水平均较模型组低(P<0.05),以附子理中汤大剂量组明显(P<0.01);模型组大鼠血清IL-10水平较对照组降低(P<0.05),各药物干预组血清IL-10较模型组升高(P<0.05)。结论附子理中汤能降低D-IBS大鼠血清TNF-α水平,提高大鼠血清IL-10水平,这可能是附子理中汤对D-IBS大鼠的具体作用靶点,具体机制可能是通过调节血清促炎因子与抗炎因子水平,调整促炎因子与抗炎因子平衡,从而达到治疗效果。
Objective： To research the impact of Fuzilizhong Decoction on TNF-α and IL-10 in rats serum with diarrhea-predominant irritable bowel syndrome（D-IBS） and to preliminary explore the molecular mechanism of Fuzilizhong Decoction in the treatment of D-IBS.Methods： Rats were randomized into 5 groups： control group,model group,dicetel group,Fuzilizhong Decoction with low-dose group and Fuzilizhong Decoction with high-dose group with 10 rats in each group.The models with D-IBS were established by intragastric administration with senna leaves and restraint stress.2 weeks later,different treatments were taken for different groups.The expressions of TNF-α and IL-10 in the serum of rats were determined by enzyme-linked immunosorbent assay（ELISA）.Results： The expressions of TNF-α in serum of model group were higher than those of the control groups.After Drug intervention,expressions of TNF-α in serum of rats were decreased obiously.In contrast.The expressions of IL-10 in serum of model group were lower than those in control groups.After Drug intervention,expressions of IL-10 in serum of rats were increased higher signifcantly（P 0.05）.Conclusion： Fuzilizhong Decoction can reduce the expression of TNF-α and increase the expression of IL-10 in serum of D-IBS rats and thereby exerts therapeutic effects against D-IBS.
