摘要
目的 探讨星形细胞瘤组织中Fas和FasL的表达及其与细胞凋亡的关系。方法 采用免疫组织化学方法和TUNEL技术检测 41例星形细胞瘤组织中Fas和FasL的表达以及肿瘤细胞的凋亡指数。结果 2 7例 ( 66% )星形细胞瘤有Fas的阳性表达 ,Fas的表达水平较低 ,3 4例 ( 83 % )的瘤细胞是Fas的表达完全丢失或阳性细胞数 <5 % ;3 2例 ( 78% )星形细胞瘤有FasL的阳性表达 ,FasL的表达水平较高 ,19例 ( 4 6% )FasL的阳性细胞数较多 (阳性细胞占肿瘤细胞 >5 % )。41例星形细胞瘤凋亡指数为 1.0 9± 0 .48。星形细胞瘤Fas和FasL阳性与否和细胞凋亡指数无明显相关性。结论 Fas和FasL在星形细胞瘤的发生和发展中可能起重要作用 ;星形细胞瘤的发生和发展与细胞凋亡异常有关 ;星形细胞瘤Fas的表达水平低下可能导致肿瘤细胞逃脱机体对其的清除作用 。
Objective\ To investigate the expression of Fas or FasL and their relationship with apoptosis in astrocytomas. Methods\ Immunohistochemical method was used to detect the expression of Fas and FasL by TUNEL technique and apoptosis index (AI) was calculated in 41 astrocytomas. Results\ Twenty-seven of the astrocytoms(66%) expressed Fas. The amount of cells positive for Fas was small. 83% astrocytomas(34 of 41) showed complete loss of Fas expression or expression in <5% of tumor cells. Thirty-two of the astrocytomas (78%) expressed FasL. In contrast, the percentage of positive is more, with 46% astrocytomas (19 of 41) having positive cells greater than 5%\, AI of astrocytomas was 1.09±0.48.The positive expression of Fas/FasL is not significantly related to apoptosis in astrocytomas. Conclusion\ Fas and FaL are important to the pathogenesis and development of astrocytomas. The development and progression is related to abnormality of apoptosis. The weak expression of Fas may contribute to astrocytomas escaping from clearance by self-immune system. Further study on the mechanism of astrocytoma apoptosis is needed.
