摘要
本文研究了以碘为显色剂 ,应用双波长分光光度法测定银杏果仁中直链淀粉和支链淀粉的含量。对显色反应和光度测定的最佳条件进行了详细的考察。根据碘 -直链淀粉和碘 -支链淀粉复合物的吸收光谱 ,选择直链淀粉的测定波长为 5 6 8nm和 42 0 nm,支链淀粉的测定波长为 5 1 2 nm和 6 48nm,依据回归方程可求出直链淀粉和支链淀粉的含量。直链淀粉的浓度在 0— 36 μg/ m L,支链淀粉在 0— 1 1 0 μg/ m L 范围内符合比耳定律。直链淀粉和支链淀粉的回收率在 98.3%至 1 0 4%之间 ,相对标准偏差小于 1 .30 %。
Dual wavelength spectrophotometry was applied to the determination of amylose and amylopectin in ginkgo biloba using iodine as coloring agent.According to the absorption spectra of the two complexes,the wavelength of I 2 amylose,568nm and 420nm,and of I 2 amylopectin,512nm and 648nm,were selected as the measuring wavelength.The content of amylose and amylopectin were figured out from the two regressive equations.Beer's law is obeyed in 0—36μg/mL for amylose and 0—110μg/mL for amylopectin.The recovery rate of both amylose and amylopectin ranged is from 98.3% to 104%.The relative standard deviation is less than 1.30%.
