摘要
目的探索从冰川地区分离和培养低温菌噬菌体,并对其特征进行研究。方法利用"双层平板法"从明永冰川地区分离得到一株蜡样芽孢杆菌烈性低温噬菌体(命名为MYPB41-22),并对其生物学特征进行研究。结果宿主菌MYB41-22鉴定为Bacillus cereus菌株。噬菌体MYPB41-22在电镜下呈长六棱柱状,具一短直尾和一"衣领"状结构,无鞘。"衣领"状结构由对称的2~4个短棒状结构组成,头部具多面体立体对称结构,长55 nm,宽40 nm;尾管长约40 nm;4℃时具侵染活性,在4~37℃均可产生边缘清晰透明的噬菌斑,最佳感染温度为15℃,最适感染pH为8,最适感染复数0.1,对氯仿不敏感,基因组为双链DNA,大小约20kb。结论衣壳蛋白分析表明MYPB41-22在52 kDa处主条带含量较高,推测可能为其重要的结构蛋白。
Objective To explore the characteristics of a low-temperature bacteriophage isolated from alpine glacier region.Methods A low temperature phage of Bacillus cereus named MYPB41-22 was isolated from Mingyong glacier region by "double plate method" and its biological characteristics were studied.Results Its host bacterium MYB41-22 was identified as B.cereus.The MYPB41-22 was in a shape of long and hexagonal prism,with a short straight tail and a "collar"-like structure with no sheath under electron microscope.The "collar"-like structure was composed of 2 to 4 symmetrical short rods,and its head was symmetrical polyhedral three-dimensional structure,55 nm long and 40 nm wide;the tail pipe was about 40 nm long.The bacteriophage could produce transparent plaques with clear and sharp edges in a temperature range of 4 to 37 ℃,and showed good infective activity at 4 ℃;The optimum temperature of infection was 15 ℃,the optimal pH of infection was 8,and the optimum multiplicity of infection was 0.1.It was insensitive to chloroform,and its genome was consisted of double-stranded DNA with the size of about 20 kb.Conclusion Capsid protein analysis showed a higher level of major protein band at 52 kDa,which was presumed to be a critical structural protein of MYPB41-22.
