摘要
目的研究Wnt信号通路的核心因子β连环蛋白(β-catenin)在大鼠酒精性肝纤维化(AHF)中的表达及意义。方法采用白酒辅以玉米油、吡唑混合食料灌胃的方法制备大鼠AHF模型,于造模12 w后,采用Masson染色法观察肝组织纤维化程度,采用Elisa法检测大鼠血清层粘连蛋白(LN)、Ⅲ型胶原前蛋白(PⅢP)水平,逆转录聚合酶链反应(RT-PCR)检测大鼠肝组织β-catenin mRNA的表达。结果正常肝组织中有少量β-catenin表达,12 w末模型组大鼠肝组织中β-catenin表达明显升高。β-catenin的表达量与肝组织纤维化程度,以及血清LN、PⅢP水平呈正相关(r=0.926,0.893,0.902,P<0.01)。结论 Wnt信号通路在AHF发生发展过程中可能发挥了重要的作用。
Objective To explore the expression and significance of β-actenin(the key factor of Wnt signaling pathway) in rat models of alcoholic hepatic fibrosis.Methods 60 Wistar rats were divided into two groups at random: normal group(n=30),model group(n=30).Rat model of alcohol hepatic fibrosis was induced by perfusing stomach with the mixture of liquor,pyrazol and corn oil.After 12 weeks intervention,variety index of hepatic fibrosis and expression of β-catenin mRNA in liver tissue were observed.Results Compared with normal rats,the degree of hepatic fibrosis,the level of PⅢP,LN in blood serum and the expression of β-catenin mRNA were increased in model rats.The expression of β-catenin and the degree of hepatic fibrosis,the level of PⅢP,LN in blood serum were positively correlative(r=0.926,0.893,0.902,P0.01).Conclusions Wnt signaling pathway may be involved in the genesis of alcoholic hepatic fibrosis and be closely related to the development of alcoholic hepatic fibrosis.
