摘要
目的:探讨膳食多不饱和脂肪酸(PUFA)改善糖尿病孕鼠脂肪细胞中胰岛素抵抗的可能机制。方法:利用链脲佐菌素(STZ)建立妊娠期糖尿病孕鼠模型,取30只模型鼠随机分成对照组、干预组1(低热卡饮食)和干预组2(低热卡饮食+PUFA),每组10只。分离孕鼠脂肪细胞粗膜、内膜、外膜;利用D-(U-14C)-葡萄糖分别测定其葡萄糖转载体(GLUT)相对含量。结果:干预组1和干预组2孕鼠脂肪细胞粗膜、内膜、外膜的GLUT含量均高于对照组(P<0.01),干预组2孕鼠脂肪细胞粗膜、内膜、外膜的GLUT含量高于干预组1(P<0.01)。结论:PUFA饮食增加妊娠期糖尿病孕鼠脂肪细胞的GLUT,PUFA可能是改善胰岛素抵抗的环节之一。
Objective:To explore the possible mechanism of improved insulin resistance in the rat model of GDM after the intervention of dietary PUFA. Methods:The rat model of GDM was established by strepto- zotocin(STZ). Thirty of them were used and randomized evenly divided into three groups: control group ( STZ, n = 10), Group 1 ( STZ + low calorie, n = 10) and Group 2 ( STZ + low calorie + PUFA, n = 10). Crude membrane, plasma membrane and intracellular membrane fractions of adipocyte were isolated by different speed of centrifugations. The relative amount of GLUT in membrane fractions was detected by D-E U- 14C]-glucose radioactivity. Results: The amount of GLUT in membrane fractions in Group 1 and Group 2 were significantly higher than that in Control Group (P〈0.01) ,while GLUT in Group 2 was markedly higher than Group 1. Conclusions:PUFA diet increase the amount of GLUT in adipocyte,which may be a segment of ameliorating insulin resistance in GDM rats.
