一株土壤源高产纤维素酶芽孢杆菌的分离与鉴定 被引量:3

摘要 为了获得产纤维素酶的芽孢杆菌,从河南省不同地方采集玉米地、秸杆垛的土壤样品58份,利用刚果红平板法分离产纤维素酶的芽孢杆菌,采用3,5-二硝基水杨酸法测定纤维素酶活,结合菌落特征、显微形态、生理生化试验和16SrDNA序列进行鉴定。结果表明,从样品中分离出的42株产纤维素酶菌株中筛选出一株产纤维素酶较高的菌株B.LY02,纤维素酶活力可达0.5351U/mL。该菌株呈短杆状,革兰氏染色阳性,能形成芽孢。基于16SrDNA序列同源性比较分析表明该菌株与Bacillus licheniformis strain CICC10095的亲缘关系最近,基因序列的同源性为96.5%,因此鉴定该菌株为地衣芽孢杆菌。 In order to obtain the high yield cellulase bacillus,58soil samples from different areas in Henan province were collected,the bacillus producing cellulase was isolated by Congo red medium,and the cellulase activity was detected by 3,5-dinitro salicylic acid method.The high cellulase producing bacillus were screened and identified by the colony morphology,microscopic morphology in combination with the physiological and biochemical feature and 16SrDNA sequence.The results showed that the strain B.LY02 with the highest cellulase activity of 0.5351U.mL-1 was obtained,and it was short rod,forming spores,gram strain positive,and had the ability of fermenting many sugars.The 16SrDNA sequence homology comparative analysis showed that it belonged to the genus Bacillus and most closery related to Bacillus licheniformis strain CICC10095with 96.5%sequence simmarity.So the strain Bacillus LY02was identified to be Bacillus licheniformis.
